The Transportation Specialist skill allows you to fulton extract vehicles, shipping containers, and other large items. It's a good thing to get early in the game because it allows you to stock Mother Base with good equipment, and allows you to use the vehicles later on.
You want to start Mission 10, and rescue the two prisoners. That's basically all you have to do! The entire rescue takes around 5 minutes and is very easy. I recommend you take D-Dog as a buddy, because he is really helpful with sniffing out guards. Just make sure you get to the prisoners as quickly as possible, because after about 6 minutes, they will either die or be executed by the guards.
