For newcomers Rome is very small (similar to Paris) and many of the main sites can be done in a day. You will need longer for the Vatican, Sistine Chapel which will take a day on their own.
Best times to go are Spring/Autumn to avoid the heat and crowd in the Summer and the cold and closed attractions in the Winter.
